Alzheimer's Disease (AD) continues to be a significant worldwide health concern, requiring an immediate search for efficacious therapeutic interventions. This extensive review investigates the possibility of Andrographolide as an innovative therapeutic intervention for neurodegeneration associated with Alzheimer's disease. The fundamental basis of the compound's therapeutic potential lies in its diverse array of neuroprotective actions, which encompass anti-inflammatory, antioxidant, and anti-apoptotic pathways. Commencing with a comprehensive examination of the increasing incidence and consequences of Alzheimer's disease, the review emphasises the dire necessity for developments in therapeutic approaches. Andrographolide, which is obtained from Andrographis paniculata, has shown promise as a contender due to its effectiveness in ameliorating critical features of Alzheimer's disease, as evidenced by preclinical investigations. The review provides an in-depth analysis of the chemical features of Andrographolide, including pertinent pharmacological aspects and information on its sources and extraction techniques. An in-depth analysis of the neuroprotective mechanisms of Andrographolide ensues, yielding valuable insights into the ways in which it regulates the complex processes that are associated with neurodegeneration in Alzheimer's disease. A thorough examination of preclinical and clinical investigations provides an all-encompassing assessment of the compound's effects on both human subjects and Alzheimer's models. By doing a comparison analysis with established treatments, the distinctive benefits and drawbacks of Andrographolide become apparent, establishing its potential as a disease-modifying drug. The paper addresses challenges encountered in research and constraints in translation. It establishes a foundation for future directions, methodological improvements, and the identification of areas where the present literature lacks.
